ABSTRACT

Abstract Bacterial leaf blight (BLB) is one of the major rice diseases in Malaysia. This disease causes substantial yield loss as high as 70%. Development of rice varieties which inherited BLB resistant traits is a crucial approach to promote and sustain rice industry in Malaysia. Hence, this study aims were to enhance BLB disease resistant characters of high yielding commercial variety MR219 through backcross breeding approach with supporting tool of marker-assisted selection (MAS). Broad spectrum BLB resistance gene, Xa7 from donor parent IRBB7 were introgressed into the susceptible MR219 (recurrent parent) using two flanking markers ID7 and ID15. At BC3F4, we managed to generate 19 introgressed lines with homozygous Xa7 gene and showed resistant characteristics as donor parent when it was challenged with Xanthomonas oryzae pv. oryzae through artificial inoculation. Recurrent parent MR219 and control variety, MR263 were found to be severely infected by the disease. The improved lines exhibited similar morphological and yield performance characters as to the elite variety, MR219. Two lines, PB-2-107 and PB-2-34 were chosen to be potential lines because of their outstanding performances compared to parent, MR219. This study demonstrates a success story of MAS application in development of improved disease resistance lines of rice against BLB disease.

ABSTRACT

Abstract The life cycle of stink bug, Glyphepomis dubia and the development of two egg parasitoids (Telenomus podisi and Trissolcus basalis) were studied at the Federal University of Maranhão, at 26 ± 2oC, relative humidity (RH) of 60 ± 10% and 12h photophase. Individuals used in the study were collected from seven rice fields located around the municipality of Arari, Maranhão, Brazil, and maintained in greenhouse and laboratory for the life cycle studies. From egg to adult, G. dubia took 35.2 days to complete the life cycle. The oviposition period was 37 days, with egg masses of about 12 eggs each and viability of 93.1%. Longevity was 53 and 65 days for females and males, respectively. The egg parasitoids Te. podisi and Tr. basalis parasitized and developed in G. dubia eggs; however, the biological characteristics of Tr. basalis were affected. Emergence of the parasitoids was higher for Te. podisi (83.5%) compared to the records for Tr. basalis (50.4%). Therefore, G. dubia may potentially achieve a pest status and Te. podisi is a promising biological control agent for G. dubia management in Brazil due to its higher longevity and better reproductive parameters.

ABSTRACT

Resistance to acetolactate synthase (ALS) inhibitors have increased recently in South Brazil where the major weeds of flooded rice (barnyardgrass and weedy rice) have evolved resistance to imazapyr+imazapic. The aim of this research was to evaluate a growth medium for tissue regeneration of tillers in barnyardgrass, as well as an agar-based bioassays test (also from tillers) to detect susceptible and resistant biotypes of weedy rice and barnyardgrass to imazapyr+imazapic in vitro. Greenhouse experiments were conducted to detect ALS-resistant (R) and susceptible (S) weedy rice and barnyardgrass biotypes, and bioassays were carried out to evaluate an adequate growth medium for barnyardgrass tiller regeneration and determine the concentration of herbicide to distinguish R and S plants. The culture medium that provided a suitable barnyardgrass growth was MS 50% with the addition of benzylamino-purine. The tissue regeneration in vitro with the growth medium containing imazapyr+imazapic allowed to discriminate between R and S barnyardgrass and weedy rice plants. The concentration required for satisfactory control of susceptible barnyardgrass and weedy rice explants grown in vitro was 0.9 µM and 1.3 µM of imazapyr+imazapic herbicide, respectively. The bioassay in vitro using tiller regeneration provides an opportunity to predict effectively imazapyr+imazapic resistance in barnyardgrass and weedy rice.

ABSTRACT

The ascomycete fungal pathogen Ustilaginoidea virens (Cooke) Takahshi cause false smut in rice and considerable yield loss. In this study, we collected isolates of U. virens from the rice growing ecosystems of Karnataka and characterized for cultural, morphological and molecular characters. The isolates of the fungus on Potato Sucrose Agar media exhibited distinct colonies with colony growth ranging from 21.50 mm (Uv-20) to 70.00 mm (Uv-15). The colony colour appeared as whitish to yellowish with varied growth pattern from flat, raised flat to fluffy and raised fluffy colonies with sectoring in Uv-1, Uv-3, Uv-6 and Uv-9 isolates. The isolates of U. virens also showed variation in the morphology of spores, where the conidia were globose, irregularly round to elliptical and warty on the surface with spore radius ranging from 2.91 to 5.36 ?m. The scanning electron microscopy revealed hyaline globose to irregularly rounded ornamented chlamydospores with prominent spines. Besides cultural and morphological characters, molecular identification of false smut isolates was confirmed through ITS sequencing which showed 91 to 99 per cent identity with U. virens in NCBI-BLAST analysis. Dendrogram constructed using ITS sequence data broadly separated the isolates into two major clusters with divergence among clusters. This ITS (internal transcribed spacer) sequencing of isolates should help better understanding of the phylogenetic relationships among these isolates.

ABSTRACT

Salt stress is a major abiotic stress that limits rice productivity worldwide including India. As modern rice varieties are salt sensitive, infusing salt tolerance through breeding is a viable farmer-friendly approach. Breeding salt tolerant rice varieties has been slow due to complexity of the trait and high Genotype x Environment interaction in the salt affected field. Selection practiced in such a situation using conventional selection method, in the presence of competition, would be misleading. On the otherhand, selection is effective in the absence of competition in honeycomb selection and counteracts the disturbing effects of competition on effectiveness of selection. The present study was conducted to test the efficiency of honeycomb selection design in early generation of a rice cross to study genetics of yield and practice selection. All the characters studied in both the design showed non normal distribution except for panicle length in CSD. All the characters studied in both the design had lower coefficient of variation, high mean and high standard deviation in the HSD compared to CSD. Large number of genes with duplicate epistasis governs days to flowering whereas panicle length and single plant yield are governed by few number of genes with complimentary epistasis. Twenty nine F2 plants each in CSD and HSD were selected based on mean and plant index (SPY), respectively. Plants selected in HSD recorded higher percentage of increase over base population compared to CSD and found HSD to be superior to CSD because of enhanced phenotypic expression in the former by eliminating confounding effects of negative correlation between yielding and competitive ability.

ABSTRACT

Acute lung injury (ALI), during the progression of infectious shock, leads to systemic inflammatory response syndromewith increased pulmonary capillary membrane permeability due to pulmonary inflammation and uncontrolled inflammatoryresponses. It may cause fatality in patients. Here, we evaluated the protective effect of breviscapine on ALI in rats withinfectious shock. Sprague-Dawley (SD) rats were assigned into Sham, model [lipopolysaccharide (LPS) group], andbreviscapine treatment groups (LPS + breviscapine group) and weighed. The lung coefficient, and the wet-to-dry weightratio (W/D) and moisture content of lung tissues were calculated. The pathological changes of the lung tissues were detectedusing hematoxylin-eosin (HE) staining, and the protein expressions of interleukin-1 beta (IL-1?), IL-6, and tumor necrosisfactor-alpha (TNF-?) were determined by enzyme-linked immunosorbent assay. Western blotting was conducted to measurethe protein expressions of toll-like receptor-9 (TLR-9) and nuclear factor-kappa B (NF-?B) (p65). Compared with LPSgroup, breviscapine significantly lowered the lung coefficient and the W/D and moisture content of lung tissues, relieved thepathological changes of lung tissues, reduced the protein expression levels of IL-1?, IL-6, and TNF-?, weakened theactivation of NF-?B (p65) in lung tissues, and repressed the protein expressions of TLR-9 and NF-?B (p65).

ABSTRACT

This study aimed to verify the efficiency of multilines in reducing blast progress and their potential benefits to phenotypic stability in rice. The experiments were conducted in the 2016/17 and 2017/18 agricultural years. A randomized block design was performed with three replications, evaluating 12 lines and a multiline, which consisted of five lines from the Cultivation and Use Value (CUV) test. The multiline presented an estimated grain yield above the average of experiments of around seven bags ha-1 and superior performance in early flowering, justifying the high phenotypic stability for these characters. In this case, the line selection for composing the multiline was favorable and efficient, highlighted by a higher agronomic performance than most lines of the CUV test. The multiline is an adequate strategy to provide higher phenotypic stability and reduce blast progress in the field.

ABSTRACT

The choice of cultivars adapted to different regions and the determination of the best sowing dates are indispensable tools for crop development. This allows the installation of the crop in times favorable to its development, as well as cultivars that manage to reach their maximum yield potential. The objective of this study was to determine the best sowing dates and the cultivars that can adapt to the low-altitude Cerrado region and have the best development and yield efficiency. The experiment was conducted on a typical clayey dystrophic Red Latosol soil. The experimental design was in randomized blocks in a factorial scheme, comprised by four sowing dates during the spring/summer season (October, November, December, and February) and in each sowing date there were eight upland rice cultivars (BRS Esmeralda, ANa 6005, ANa 5015, IPR 117, IAC 203, IAC 500, ANa 7211 and BRSGO Serra Dourada) with four replicates. Sowing in October and November benefited the aerial dry mass and the spikelets fertility however, sowing in November provided higher plant height, which caused lodging in plants. Sowing in December caused the incidence of scald, mainly affecting the cultivar ANa 7211. For all cultivars, sowing in October favored the yield efficiency of upland rice in the low-altitude Cerrado, followed by sowing in November. The cultivars which demonstrated higher yield efficiency in the conditions of the region were BRS Esmeralda and ANa 5015.

ABSTRACT

ABSTRACT: The purpose of this study was to evaluate the contribution of the inoculation of the strain Azospirillum MAY12 to the development of rice plants of the cultivar BRS Esmeralda. A completely randomized experiment was performed in a greenhouse, with 8 treatments and 6 repetitions, namely, commercial inoculant (CI) containing strains of Azospirillum brasilense ABV5 and ABV6 (T1), MAY12 (T2), nitrogen control with 50 mg of N or full dose (T3), CI + 30% of N (percentage referring to full dose) (T4), CI + 60% N (T5), MAY12 + 30% N (T6), MAY12 + 60% N (T7) and absolute control (T8). There was a significant increase in the fresh and dry mass of the shoot and roots, height, root volume, and the number of tillers and leaves of plants with the MAY12 inoculation (with and without N added) compared to T8. Treatments containing MAY12 + N increased the total dry mass by ~920% compared to T8, matching the T3 yield. The inoculation of MAY12 in isolation was as efficient as the application of the full dose of N, resulting in an increase of ~115% in height and 373% in root volume of the plants compared to T8. The MAY12 strain demonstrated good performance in the development of the cultivar, which was comparable to the CI in the variables tested. Therefore, inoculation with the MAY12 strain is promising for the partial reduction of nitrogen fertilizers in upland rice cultivation.

ABSTRACT

ABSTRACT: Knowledge of the effect of the seedborne inoculum is important for knowing the level of tolerance of the pathogen by the seed. This research evaluated the effect of the incidence of the fungus Microdochium albescens on the physiological quality of the seeds of different cultivars of irrigated rice. The study was carried out in the seed testing laboratory (STL), phytopathology laboratory and in greenhouse, located at the Center of Agroveterinary Sciences of the Santa Catarina State University, Lages, SC, Brazil. Seeds from six irrigated rice cultivars produced in the Alto Vale do Itajaí Region in the 2016/17 harvest, were used. The lots were submitted to the seed health test, identifying four lots per cultivar with a natural incidence of M. albescens, with two lots of each cultivar with an incidence greater than 40% and two lots equal or less than 40%, totaling 24 seed lots. The following evaluations were carried out on the lots: germination, accelerated aging test, emergence in a greenhouse at 14 days, emergence speed index (ESI), emergence speed (ES), shoot length, root length and fresh and dry mass. There was a significant interaction between cultivar and level of incidence only in the variables germination, vigor and root length. Fresh mass, shoot length and ESI were not affected, regardless of cultivar and incidence of fungus in the seed. The irrigated rice cultivars SCS118 Marquês and SCSBRS Tio Taka are susceptible to a high incidence (> 40%) of the fungus M. allbescens.

ABSTRACT

ABSTRACT The fall armyworm, Spodoptera frugiperda (J. E. Smith) (Lepidoptera: Noctuidae), is an important rice pest which consumes leaf area during all phases of plant development. The objective of this study was to identify rice genotypes that provide antixenosis and antibiosis to fall armyworm. Antixenosis was investigated by measuring non-preference in free-choice and no-choice tests while antibiosis was determined by measuring biological parameters and nutritional indices of S. frugiperda in twelve rice genotypes. The genotype Miúdo Branco showed antixenosis to S. frugiperda, whereas the genotypes IR 64 and Bacaba Branco showed antibiosis to S. frugiperda by affecting insect development and prolonging insect life cycle. Insects that fed on Bacaba Branco genotype demonstrated the lowest nutritional indices. Since Miúdo Branco, IR 64 and Bacaba Branco showed moderate resistance, rice producers could use these genotypes as part of a control strategy for S. frugiperda.

ABSTRACT

Abstract Kriging is a method that estimates values in places not sampled from different interpolators, therefore, widely used to predict the spatial distribution of organisms. However, the different interpolators may vary in performance depending on the organism under study or the area evaluated. The aimed study to compare the ordinary kriging and inverse of distance weighted interpolation methods, applied to the spatial distribution of population density of Tibraca limbativentris in irrigated rice. This study was carried out in Santa Maria, RS, Brazil, in two fields with areas of 1.3 ha and 6.2 ha, respectively. Seven evaluations of the population density of T. limbativentris were carried out, corresponding to the period from sowing to maturation. In these areas the adults of T. limbativentris were quantified and the sum used for the statistical and geostatistical analysis. The sample population of T. limbativentris was submitted to different semivariograms, which were selected through cross-validation. The sample population of T. limbativentris was submitted to different semivariograms, selected by means of cross-validation. Once selected, semivariograms were used in both tested interpolation methods. From the results it was concluded that the ordinary kriging interpolation method performed better in all evaluations performed in both areas. Therefore, we recommend its use for estimating the population density and spatial distribution of T. limbativentris in the irrigated rice throughout the crop phenology. Using appropriate interpolation methods, localized management can be used, reducing costs for controlling this pest and increasing the sustainability of the environment.

ABSTRACT

ABSTRACT: Over post-harvest steps of rice, from pre-cleaning to processing, a large amount of by-product is generated. Some of these by-products, due to their high starch and fiber content can be used in ethanol production. The objective was to evaluate the effect of enzymatic hydrolysis conditions on the production of reducing sugars, from pre-cleaning residue and type III paddy rice, as well as the effect of the pre-treatment of its fibers, targeting the use of these residues in ethanol fuel production. The proximate analysis was performed, followed by the pre-treatment of samples. Enzymatic hydrolysis was conducted in two ways: using one enzyme at a time or applying them simultaneously. The starch content was 41.18 and 53.41%; the fibers were 30.44 and 23.39%, of which 6.53 and 4.41% were lignin, for the pre-cleaning residue and paddy rice, respectively. Alkaline pre-treatment reduce lignin content by 47.94 and 18.23% for the pre-cleaning residue and type III paddy rice, respectively. Hydrolysis efficiency was 22.61 and 15.32% for the cellulase enzyme, and 82.18 and 87.07% for the amylolytic enzymes in the pre-cleaning residue and type III paddy rice, respectively. The hydrolysis with the separated enzymes presented higher reducing sugar yields. Therefore, the pre-cleaning residue and type III paddy rice can be used for ethanol production by its enzymatic hydrolysis, aiming to add value and to increase the sustainability of the rice production chain.

ABSTRACT

Mesocotyl elongation is a key trait influencing seedling emergence and establishment in direct-seeding rice cultivation. The phytohormone gibberellin (GA) has positive effects on mesocotyl elongation in rice. However, the physiological and molecular basis underlying the regulation of mesocotyl elongation mediated by GA priming under deep-sowing conditions remains largely unclear. In the present study, we performed a physiological and comprehensive transcriptomic analysis of the function of GA priming in mesocotyl elongation and seedling emergence using a direct-seeding

ABSTRACT

Farmers from the district of São Pedro de Joselândia, belonging to the municipality of Barão de Melgaço, Pantanal of Mato Grosso, have been cultivating for some time two rice plant materials and three types of beans that, according to them, are not commercial. For this reason, the objective of this work was to test the originality of the materials by means of genetic and morphological characterization in order to identify whether the nature of the subsamples would be effectively traditional or whether they are varieties already known in the market. Morphological analyzes of the cultivated plants (phenotypic characteristics, cycle and flowering) and seed of the rice subsamples, hereinafter called Agulhinha Vermelho and Branquinho and of the Amarelinho, Rosinha and Roxinho subsamples were performed. Also, the evaluation of distribution of comparative genetic variability with other materials of the same species, already cataloged, was determined. The subsamples were characterized as traditional varieties different from those already cataloged and they are not found in the Nuclear Collection of Rice and Beans of the Brazilian Agricultural Research Corporation (Embrapa). They have distinct morphological characteristics, being potential species for use in the market of new technologies. This should guarantee to the residents of that community that they are breeders of specific materials of cultural heritage adapted to the local conditions.

ABSTRACT

The bioavailability of a nutrient in a food serves to determine the nutritional quality for human consumption. Rice is one of the most important foods in diet due to its caloric and nutritional contribution. The objective of this study was to analyze the in vitro bioavailability of zinc in rice by ultraviolet-visible spectrophotometry and flame atomic absorption spectrometry and its relationship with phytic acid content. The percentage of zinc bioaccessibility with respect to phytic acid percentage, showed an inverse proportional logarithmic relationship (r= −0.669; p<0.05). The percentage values of zinc bioavailability and phytic acid in the evaluated rice varieties had a range of 1.9-8.7% and 0.039-0.946%, respectively. Phytic acid affected the bioaccessibility of zinc and was not linked to the total concentration of zinc present in the rice lines evaluated. The techniques implemented to quantify zinc dialyzed presented significant differences. It was shown that the ultraviolet-visible technique was not suitable for this type of assay.

ABSTRACT

Aim: To identify bacterial leaf blight resistant genes in some rice varieties so that these resistant varieties can be used as a good source of donor for BLB resistant genes in genetic enhancement program.Methodology: A total of sixty-one rice genotypes including resistant and susceptible checks were screened in field condition by artificial inoculation using IX020 strain of Xoo for two years (Kharif 2016 and 2017). These varieties were also genotyped for seven SSR markers tagged with major BLB resistant genes, i.e., Xa4, xa5, xa13 and Xa7. Results: In artificial screening, significant disease development was recorded and the varieties were categorized using disease scoring scale of IRRI, 1996 where seven cultivars exhibited resistance, while twenty-seven were found to be moderately resistant. In genotyping, there was distinct difference in banding position for resistant and susceptible genotypes. Genotypes having resistant disease reaction, carrying BLB resistant genes were identified. Interpretation: Genotypes IR-64, IR-68144-2b-2-2-3-1-127, Ratna, Surjamukhi, Kalinga-2, Azucena and Zheshan-2 expressed bands of RM markers closely linked to Xa4, xa5, xa13 and Xa7 BLB resistant genes and field testing also confirmed resistant host reaction against pathogens.
